500hp+ would probably require ECU tune, headers, cam shaft, supercharger, and maybe some exhaust and intake work. My guess is somewhere between 15K and 18K, depending on the base model.

Comparing the CLK500/550/55 with the route of going with a C32/SLK32 with $10K (parts + labor) to see if it would be easier to obtain 500+ hp.

Kleeman has supercharger, ecu, and cam packages for the M113 (500/55) but not for the 550. I looked into a supercharger and different tune(required) once for my 550. It was way too expensive, I would have to retune annually to get emissions testing, and I would be a "one off" setup.

For 500hp you'd have to supercharge and tune a 55 and on a 500 supercharge, tune, cams, headers:

You could get kind of near with the C32 or SLK32 but once you max it out you either need to go to engine swap, turbo's or centrifugal supercharger... expensive and difficult. Starting with a larger displacement engine would be better. Can you do all the labor yourself? E55 of course would be the easiest way to go.

Supercharged CLK500 & ML500
+1 on what the guys said above. I have almost all the mods you listed and just the parts alone will go for around $10K. It would be easier to get a W211 E55 or buy a CLK that has those mods installed.
