Is it possible to damage transmission using the sport shifters
Im new to shifting with the rocker buttons in the back of the steering column so I am wondering if it possible to damage the transmission if I shift wrong on my 06 ML350. I did search but did not find an answer, sorry for the newbie question but I have to ask.

no. it won't let you overrev the car. I guess if you incessantly shifted up and down every 2 seconds it would cause some damage, but there is no reason to do so, so you'll be fine.

Anyone know if the system will upshift above the selected gear if you're being pushed down a hill in low gear? I know it will simply ignore inputs that downshift too far.

Out Of Control!!
My GL will upshift when I tap the paddle when I am towing. (I have not tried it down hill, but do it on flats to drop engine RPM.

Long downhill while towing, you manually select 2nd gear for engine braking. Your speed continues to climb even with the engine braking to the point where the car should upshift but you've manually held it to 2nd. Will it go to 3rd (presumably) to prevent overrevving? This would be a rare condition to run into but one that I've been curious of. If it doesn't upshift automatically that's the only condition I can think of that might cause any damage.

Yes.
One thing you learn driving a heavy truck is to go down hills in the same gear you would use to go up. Prevents over-use of brakes and hence brake fade. I use the same rule when towing my boat (although I have never had to hold second gear for long periods, even driving through the mountains).
