Exhaust switches off after car not being in use.

I have a 2016 C63s and I usually leave the exhaust on for a nice rumble when I start the car. Lately I find if I leave the car for a few hours or overnight, when I switch the engine on the exhaust has switched off, this makes for a disappointing start sound in comfort mode.

Is there any setting or reason why the exhaust would switch off when the car hasn't been running for a few hours?

That's standard behavior. The drive mode reverts back to Comfort which has the exhaust valves closed by default when the engine is shut off. Not sure why you only have noticed this lately, but that's how it works. It's due to noise and fuel economy regulations. There are some third-party modules that can keep the exhaust valves open at all times, automatically turn off ECO start/stop and engage Individual automatically upon start up. There's no setting out of the box that lets you start the car with the last settings. The 2019+ models have what's called Emotion Start. By holding one of the shift paddles the engine starts with the valves open. For the PFL you can turn on the ignition, then select the drive mode you want and/or open the exhaust valves, and then start the engine.

That's wierd because I have had the car for 6 months and it always starts with the exhaust on if that's how I left it when I turned it off.

In that case, one of the previous owners likely installed an ASR module. As far as I know they come with a remote to control the valve settings, and can remember the previous setting. Here's a demonstration video.
