I assume that you're asking about problem areas to investigate with the car.
I have an '03 E500 with 27K on it. It's been reliable, actually, but maybe the mileage is still too low for all the reliability problems that others report with '03 E500s.
Check the service records on the car you are considering, if available, to see if the SBC recalls have been performed on the car. There were two, I believe. Also check to see, if possible, whether the audio gateway and/or firmware have been updated, and whether the engine mounts have been replaced. Those are other problem areas with the '03.

Well few things you should look as some of the other have said the same...
1) Def. buy an extended warranty, and make sure research it thru so it covers everything
2) Now that you are outta warranty, make sure extended warranty does cover all the TSBs
Most of your problems likely have been worked out by now...but still get all the details from any MB dealer on that particular car, take it in to them for an inspection before buying, costs about $125 or so but it'll be money well spent
Mine E500 has been a great ride, except the all the electrical issues had to go thru.
-Stereo(replaced got a new one, but this one still is spotty)
-Heater
-Heated seats(both front)
-Rear window defogger
-Headlight fogging up
etc etc..
so def. look into what has been done to it so far.
