For '06 as far as I know the Appearance/Sport packages have: 18" multi-spoke wheels, lowered "Sport" Suspension, darker/black-ish wood inside, different front bumper and side skirts, dual exhaust tips, white face gauges & Cluster, blue tinted windows, LED rear lights, different front Grille.
Some of the simple appearance stuff is just preference, but the most important things that would cost some $$ to duplicate would be the Suspension, Rims, Bumpers if you like the look more, etc.

The sport package is more appearance than anything. The important options are:
Airmatic
Parktronic
Leather
Rear Sunshade / Side window shades
Heated-Ventilated Seats
Drive-dynamic seats
HK Sound

There is a difference between appearance/sport package and the AMG sport package. I don't know the exact differences off the top of my head, but I can try:
appearance:
blue tint
led tail lights
bixenon
appearance body kit
18" 10 spoke rims (staggered i think)
white gauges
black wood
airmatic (maybe but not sure)
AMG package:
AMG body kit
Dual exhaust
18" 5-spoke rims (staggered)
lowered, stiffer suspension
others...

But the AMG sport package does mean a stiffer and lower (and conventional spring type) suspension which I personally like. And it means you can easily tune it with aftermarket components later on in its life.

I have driven E350 non sport and sport package cars as loaners when my car is in for service, and the sport drives SOOO much better. Feels much better on the road and handling is night and day. The non sport really feels heavy and sluggish.

I agree. Those options can't be added later. The current version of the E350 doesn't have airmatic, ventilated seats, or drive-dynamic seats as available options anymore.....

As Timeless says, there is a difference. I also immediately took off the OEM Conti All Season tires my E350 came with, and replaced them with Michelin PS2. A huge difference there, too.
On the 2006, you could always get an aftermarket suspension, like Bilstein PSS coilover system. And then new wheels and tires. Add a performance exhaust system. Then you'll have even better than the OEM sports pkg.

The Appearance/Sport package cars seem really easy to find for me But I would say, either go for the Appearance/Sport or AMG/Sport as the regular edition won't be much fun to drive, or look at (comparitavely speaking) in stock form.
Also, I'd recommend if you're going to have the car out of Warranty, don't get one with an Airmatic suspension, as if it breaks it'll cost some $$.

You'll know the AMG Package as it'll look like an AMG, it took me a while to find mine but I loved the AMG look on the 211 so much I figured it was worth it to me, they're quite "rare" so people usually charge more for them.
