Just picked up a "new to me" CPO, 2010 E350 with 21600 miles. What a fantastic car so far.
I've already done a lot of reading here and picked up a lot of information. Thank you very much.
1st question, is there anything I should be looking out for?
Warranty is good until end of 2014, or 100,000 miles..

If you plan to keep it past 2014 and put less than 20K miles per year, look into extended warranty.

Congratulations! I'm finding 2010 E350, also my first MB, a rewarding experience. Had power steering connection fitting recall done that affected some 2010-2011 E's. I imagine a CPO car would have already had that done and latest command update and so on. I also had fading wood trim pieces, to which dealer replaced all wood trim. This is major in the sense it would cost a lot of money, and I was told it is not an item that is covered during the MB extended warranty - only covered during factory warranty.
I highly suggest you read the first sticky in thread started by konigstiger, who has been tremendous contributor and wealth of information.
Happy rolling on our E!

You be wise to have two documents. The data card for your card so you will know exactly the build info including engine number. That way, you can evaluate whether bulletins apply to you or not. Also, the VMI will show the service history. See what recalls were done and service or trouble issues. Congrats. You may notice fuel economy may be lower than newer models because yours is before direct injection engines. Not a worry. Just a design change.

Threads 1 and 3 are the best on this site. Have to wade through a lot, but well worth it as this is where recalls, TSB's, etc. show up.
May also want to join truedelta.com under the W212 section. The reliability section there lists specific issues people have had with the cars.
Enjoy the car and read the Operator's and COMAND Manuals throughly.
