It really only comes up on fuel economy. It has less power, more NVH, and will probably be slower than if they put the old 3.5 in there. Also, currently, the sport and luxury packages are no cost options, and I am sure the sport will be a good 1500-2000 to add to the base price.

Considering no one has driven one yet, arguing about NVH and performance are both moot. That being said:
It has the same amount of torque as the outgoing 3.5L, only at a lower RPM range. In normal driving situations, it should feel quicker than a comparable 3.5L car and seem like it is exerting itself less since it will most likely ride the boost rather than revs like the 6 cylinders do.
If the W204 and W205 C classes are any example it will be quieter than the V6 due to it not having a coarse (and IMO intrusive) exhaust note. A W204 C250 is a hell of a lot quieter than a 3.5L W204 C300. A W205 C300 is quieter than a C400 for the same reason.
I am not so sure that the fuel economy will actually be that much of an improvement over the 6 though if it is spending a lot of time with the turbo spooled.

I dont think there is any comparison between a 6cyl and a 4 when it comes to noise and vibration. It doesn't matter if I have not driven the new E class since it is the same engine as found in the w205. Haven driven that, and can compare it to the current 3.5 in the E class, and the 3.5 in my W204 that is closely related, in my opinion, the 6 is smoother and has a more pleasant exhaust and engine note.

IMO, Mercedes has not made a V6 since the W203 chassis with a decent engine or exhaust note, period - they are all grating and intrusive.

I had the M276 3.5 DI in my '13 C350, and I have it in my '15 E350 as my wife does in her '12 SLK350. I've driven the turbo 1.8 and turbo 2.0, fine engines, but not for me. My opinion of course.
I'll add that the W204 chassis as a C250 works far better than the heavier E class, from my experience, understanding that the W213 will be just a bit lighter. If you want a nice exhaust note, there's nothing like an AMG V8.
