My 2010 GL450 has in-dash DVD slot as part of the Command Head Unit. Is there also a DVD loader in the back below a seat? I keep seeing references to below-seat DVD player; I'm not sure if that's only for 2007-09 (or some 2010 option) because I don't see any DVD player under any of the seats.

It should be in back of the center console below the AC vents (if so equipped). It hinges down and open by simply pulling it down.
