Does GL550 have a stiffer suspension?
I know the tires are wider and lower profile, but is the suspension any different on the GL550 as compared to 450?

Yes, there is multi-stage AIRMatic with stiffer settings.

Are they equally soft in the comfort mode, or is the ride in 550 still harsher?

imho a lot of the stiffness would be from the larger wheels / lower profile tires

I run my GL63 in Sport Plus all the time and the ride is still reasonable

Comfort mode in a GLS550 is very smooth but the same ride in a GLS450 is a bit smoother and a bit quieter due to the tires and smaller engine. That said, the 4.7 V8 in the GLS550 is a great engine and you'll love it when you push the pedal to the floor. Test drive both before you buy.
