GLS450 random Cyl3 and Cyl6 Misfire
After being out of town for a few days, I moved the car a few feet into the garage. This morning, I cranked her up and could tell immediately something was not right and the car was running rough. Check engine illuminated (but never flashed), and the car ran rough for several miles clearly misfiring and shaking with limited power. My scanner showed random misfire code, and misfires on Cyl3 and Cyl6. I turned the car off and on at a traffic light, and voila, all was gone--including the check engine light. What in the hell could that be?
The cam phaser makes a bit of noise on cold start but is not very noticeable. No other codes on the MBII scanner besides the misfires.
My understanding is 3 and 6 are the rear-most cylinders.

Mine did the same thing a few years back. I don't recall there being a code for a specific cylinder(s), but I probably didn't use an MB specific scanner. Called the dealer and was told "that happens once in a while" or something similar. Hasn't happened since...
