How do you use the phone?
If the call is already in progress, Bluetooth must usually be disconnected using the phone's menus. Once the connection is broken, the phone is used in-hand. This is true for all MB models. You'll have to consult your phone's instructions for disconnecting Bluetooth.
Also -
For all MB models with COMAND or radio except the '07 S-Class, before a call is established (i.e., when initiating a call), press and hold the TEL button on COMAND or radio for 3 seconds to disconnect Bluetooth.
For the '07 S-Class, using the COMAND controller, select "Telephone" on COMAND, then "telephone" (on the resulting COMAND menu), and "telephone off."
I have never tried simply picking up the phone and muting the car's audio - it might not work, but if it does, it is simpler than the other steps.
For non-Bluetooth plug-in devices, do what My06clk said.

Anyway, in order to make any call private IE: turn off the car speakers, You can either unplug the BT module, OR simply turn off bluetooth on your phone. Either way it will automatically revert to handset without interrupting the call.
Since I may or may not have passengers in my car, And I like privacy when I have passengers, I have my Pebl set so that it will not automatically attach to my car when I get in. this way if I receive a call I answer it on my handset. When I am ready to go "public", I just turn on my bluetooth and the phone asks "do you want to temporarily enable bluetooth?" I say yes and it attaches to my car.
When I turn off the ignition, it automatically cuts the bluetooth back off.
The next time I enter my vehicle, I have to repeat the enabling if I want the car to answer my calls. Usually I am not in my car very long, so I keep it off.
