Can the sl55 rims fit on my w220
Front Wheels: 8.5X18, offset 25mm, tire (255/40ZR18)
Rear Wheels: 9.5X18, offset 28mm, tire (285/3ZR18)
I have a 2000 s430

I know for a fact the 285 series rear tire won't work with that offset.
Rule.
S wheels will work well on as SL but not the opposite.

The way I've always understood it is, the lower the offset number, the further the wheel will extend from the hub? So offset numbers such as ET25 and ET28 will actually have the wheels sticking out from the fenders on a W220? Or no?

The lower the offset the closer the wheel is too the hub. High offset means more space is between the hub and the rim. Thats why a low offset rims can have a larger lip, its sits back closer to the hub. High offset is was actually pushing the rim out of the fender wheel, unless you have a multi-piece rim you can counter that effect.
