I’m helping a friend diagnose a Cylinder two misfire on a 2006 S430. This issue only happens under load around 5000 RPMs at roughly 60 mph right before a shift. New plugs and wires, we’ve swapped the number 2 coil with no change. When this happens, the car runs very poorly until it is Reset via star. No other codes other than cylinder two misfire. The only other symptom would be that the car seems to run very rich at idle, strong fuel odor at the exhaust.

read your O2 sensors with SDS - it does not sound like your mixture goes "closed loop".
If cylinders run too rich, the MB ECU will shut those down to protect the cat(s).

Everything appears to be functioning within the parameters at idle. What exactly should I be looking for?
Wish people would follow through with their findings and repairs! Too many threads like this. Always new people just getting what they need and leaving. Rant done 😫
In this case the solution was selling the car as is on craigslist. My friend didn’t want to put any money into it as it had a lot of other problems as well.
