Newer is better. And for heaven's sake, get a pre-purchase inspection by a competent shop that has M-B diagnostic tools -- the laptop, not just an OBD scanner. And preferably NOT the one that's been maintaining it for the current owner.
Used cars are going for a high premium right now. I'm seeing crazy prices for W221s and a lot of other stuff. One would assume prices will come back down at some point, if you're not in a hurry. Or maybe not.

I have a very high spec, in your naming S550 (Europe kept the iconic S500 naming) - I was given a loner w203 4 pot compressor manual whilst having some work done on mine - it drove better, had more far comfortable seats and I quite liked it... only the manual box meant it wasn't as easy in traffic. If it was an auto I should have swapped it directly without even bothering to exchange cash (and this loner was an unloved high mileage dog !!!) Had I done that I'd have $30k in my pocket from maintenance, stupid road tax and insurance costs and wouldn't have spent 10 hrs a week on Merc forums trying to help others get over their mistake.
My father has had two Merc's an E320 W124 with the rare 5 speed auto option - it had awful seats with inadequate lumbar (even with the orthopedic option) and a throttle pedal that needed 6 football players to get off idle - He replaced with a E280 W211 tractor, it has awful seats. Both his have done double the miles mines done and had far less issues. Both were far more entertaining to drive, being small enough and light enough to go where you point it. The S class is like driving a Semi-Truck
The only thing an S class can have, you might aspire to is distance adaptive cruise control (they get a shiny bit of plastic on the grill covering the radar). If it hasn't got that you'd be better off walking
