2006 S55
I have the same problem on my '06 S55. Found through the forums that it was the supercharger clutch engaging, and it also screeches when initially engaged. Putting the car in Comfort mode on the tranny will reduce the problem or just go real easy in Sport mode. I've just decided to live with it.

So is this a problem to be "fixed" or is it a normal characteristic of the supercharger?

Borderline normal. From what I heard, it can be mostly fixed with a new clutch/pulley and problem should be reduced by 80+ %

After speaking with 06S55 and switching to comfort mode, the problem is almost non-existent. I am still having a tough time believing that this is something that mercedes seems to find acceptable. If I had bought this car brand new and paid sticker for it then was presented with this i'd have driven it straight back to them.

I still think it may be the pulsation damper....but am unsure of how to do a test to verify.
