Anyone know how to re-calibrate a speedometer on a 2010 GLK350 I put on new 20" RIMS and some 255/30ZR20 Accelera Phi 92Y XL - 300AAA and the speed seems off ??? Any help would be great, Thanks

The recalibration kits I have seen for 4WDs usually involve replacement gears for the take-off unit that mounts to the tranny.
Obviously nobody makes those for the GLK, although the way these things are designed it probably would throw an error code if they did.
I wonder if it can be done in the programming...

I doubt that. Usually you are supposed to keep the tire in the overall the same diameter. The car is calibrated for that diameter (ABS, Stability control, tracking control, tranny etc.)

The days of mechanical speedo cables are long gone. The sensor is a pulse generator and its output is used by the computer for all sorts of things from transmission shifting to throttle response. As such the pulse per mile figure can usually be modified either through the engineering menus, if you have navigation or through the OBDII connector. A dealer or independent MB mechanic should be able to do this for you.
If you find the vehicles engine or transmission response characteristic a little off get it done, otherwise, calculate the difference and just keep it in your head to apply it to the speed. Don't forget that the indicated miles could be 5 or 6% off.
