When did the production of MY16 start? Does anyone know whether it is different for US v German made vehicles? Is the spec much different or does it vary too much from country to country? Is the biggest difference potentially improved build quality from more cars coming off the line?

Yes, every country has quite distinct specs for C63. United States doesn't even get the Estate version, or the carbon fiber splitter/diffuser etc.
Biggest gain would be improved reliability. As far as I know, the MY16 cars don't have any new significant features over the MY15.
