After lots of tests at the 1/4 mi. strip, my fastest runs have been in D with ESP on, surprisingly. The computer shifts much faster than you, so let it do all the work.
I don't have the timeslip on me, but I'm pretty sure the fastest 1/4 mi. time was a 14.3 @ 98mph.
Here are the other methods I tried:
D, ESP off
4, ESP on
4, ESP off
Shifted from 1, ESP on
Shifted from 1, ESP off

DOH! I totally forgot to tell you what to launch at, thanks RJC.
I've found that 1600 is the highest I can launch without spinning too much off the start (and getting bogged down by ESP), and that's with 9.5" rims in the back!
