I don't know for sure since I have a cab (no speakers back there), but if they are like the center front speaker, then it tricky and yet easy.

The center speaker grill is snaps in place by little triangular tabs ("snaps" is a bit over the top, since the dash material is squishy). Basically, I just pryed it up, pushed the tabs in, and pulled it up all the way until it came out.

(Just don't blame me if it breaks something.)

i have a 04 CLK 500 and as far i know there are no rear deck speakers, there is on woofer in the rear deck on the driver side. i've popped both of the rear speaker covers off in the rear deck and there is no speaker on the passenger side just the factory woffer on the driver side, (which is now augmented by 1.6 Kilowatts of bass in the trunk.)

Remove and replace rear deck and woofer

To remove rear woofer and rear deck : 2005 CLK320 W209
Lower rear seats, pry to unsnap 2 black buttons facing the rear seat; remove C pillar screws and covers, remove speaker grills (snap up) unthread 3 roller shade nuts from trunk, pull deck forward partially, disconnect speaker wire, disconnect roller wire (on driver side), pull deck forward and free. Be careful to support the deck and the heavy roller. Release 2 woofer plastic tabs from the trunk. Lifts woofer up and out. Takes 15 minutes.
