You can supercharge a 500 for sure, but I don't think an E55 will fit. I think you need aftermarket, like Kleeman. The best thread on it here is by a guy with the screen name Kevblah. He disappeared suddenly about three years ago, but you can read his posts.

Edit: Kevwood, not Kevblah.

The E55 supercharger should fit as these are both M113 engines (the 5.5l version just has longer strokes providing the additional capacity). They also both use ME 2.8 ECUs, so that should work, too. The only issue might be that SDS can't code a CLK for the engine, or, if it does, it expects a 5.5l not a 5.0l.

Technically it's possible, but it's super complicated and expensive. Here is video
showing 5.0 m113 from CLK with installed amg supercharger (unfortunately it's only on Russian with no subtitles). They had to make custom pistons to decrease compression ration (I don't think that it's really necessary with low boost), made custom pulley, changed lots of things on the engine front cover, etc.

Interesting. I wonder why they didn't just throw in the AMG crankshaft?

I'd assume because for them it's much cheaper since they are professional Mercedes tuners and in Russia labor usually much cheaper than parts :-D

Generally easier to swap the E55 engine, loom and ECU than it is to fit just the supercharger to the 500 engine. In the US engines are a lot cheaper than all of the parts you would require.

Shouldn't even nee to swap the ECU - they both use ME 2.8.

Supercharger clutch and bypass valve operation I believe is ecu controlled and saves having the hassle of larger injector tuning and changing from the kompressor map system to the much inferior maf (for boosted applications).

It's odd. I poked around in SDS (albeit in "demo" mode which is not 100% accurate) and I found no coding for having the supercharger or not. There are some setting in EZS for engine/car but that's it. I almost wonder if the ECU just automatically detects the presence of the supercharger and reacts accordingly.

I heard somewhere that you have to define a car type (e500, e55, etc) and according to that ecu determine which equipment do you have and how to control it.

Yeah, that's what I was looking for in SDS but there are very few adaptations in the ECU coding. There are more in the EZS. But as I said, the "demo" mode is not always 100% accurate in terms of the options you seen when connected to a live car. My point is, that since both engines use ME 2.8, the original ECU should work. There will wither need to be some coding, or, it will "auto-detect" the supercharger.

I do know you need to add pins for the clutch pulley etc if you use an NA M113 loom. Not sure if that helps.

What

so would that not allow it to run or would that not be a big issue?

ECU won't be a problem, there are a number of people running Kleemann superchargers with stock NA ECUs (obviously with remapping). Are u considering Supercharging your car?
