My 2021 E450 coupe uses a wire to connect my phone to the car for Android Auto.

Other brands now offer wireless Android Auto.

Is this something that can be addressed by an over the air update, or does the car's hardware preclude wireless operation for this feature?

No way to update it. MB doesn't yet support wireless Android Auto/Apple Carplay. There are some 3rd party adapters that are coming to the market soon from what I've seen. Not sure if any are available yet.
