E450 E53 WHEEL SWAP
So I want to swap my wheels and tires with E53 wheels and tires. the wheels and tires I currently have are 19 Goodyear runflat and I want to swap them for e53 20 pirelli zero the only concern I have is that the 20inch Pirellis are stagger Tire Size: 245/35R20 (Front) 275/30R20 (Rear) Wheel Size: 20 x 8 (Front) 20 x 9 (Rear). will these work or not?

I believe they might fit with no issues, as I have an E 300 Coupe with the AMG 20inch wheels from factory. I even installed oversized tires with no issues.245/40/20 275/35/20 However, I am not sure if you need to have the wider wheel arches to accomdates the AMG wheels, as my car has the option "776 Wider wheel arch for AMG wheels".

Fitment aside, since you're going from 19" wheels to 20" wheels, does this affect speedometer and other functions due to the size change? I wouldn't mind changing over to 20" wheels at some point as well, but I don't want it to affect my driver's info.

Yes, I understand this. What I want to know is if you put different wheels on your car, can you adjust the speedometer somehow to show the correct speed? Back in the day (well...my day) transmissions had a gear for the speedometer cable that you could change out to have the speedo read properly. Now it's not so simple.

In my current C238 I could not find such a configuration.
However, I think an advanced tool like MB SDS would allow you to do so. If you own one, or have a dealer / indy do it for you.
Also, I remember the tool would allow you to choose only tire size configurations that were offered to your specific car model. (If you oversize/undersize) out of the options offered to your car model, chances are you would not find that size configuration.
