2001 E320
Transmission
2001 Mercedes E320.... Driving fine.. came to a stop.. took off... the car made a thump hesitation noise... continued to drive the car for about 200 miles.. stopped the car.. started back up.. the car made another thump hesitation and check engine light came on.. Is this the transmission?

thump....bring car to someone with code reader. sometimes auto parts store in your neighborhood will do this for free.
the codes, if detected, may give a clue as to the cause of the ailments. trying to guess this over the internet is weird.

+1 to the code reader or getting them read. These transmissions are pretty reliable. Hopefully you just need a transmission service, and it's having issues due to low fluid pressure (1st w210 I bought had this problem). Hopefully.
