With my former M273 car, setting Transmission to "M" was a big advantage due to quicker shifts. However, in my M278 there is no difference between "S" and "M" regarding shift times.
My M278 (stock) does really fly!

My car is stock and does 0-62mph (0-100km/h) in 4.6 to 4.9, depending on road condition. Measured with RACELOGIC Performance Box (10Hz GPS).

Mercedes Germany was claiming 0-62mph in 4.8 for 4matic cars with 2 person on bord. So my car is pretty much spot on.
However, I do have the Twin Turbo engine (2012-2016). Two buddies of mine have 2010-2011 5.5 N/A E550ies! One 4matic, one rwd. I was able to measure the 4matic with two person on bord and it went 5.3 for 0-62. That really surprised me, as the car feels a lot slower than the twin turbo. If one looks carefully on the data, 1st gear acceleration is similar between both engines. 2nd gear is stronger in the twin turbo. 3rd gear is even stronger. Therefore 62-125mph (100-200km/h) is a huge difference (11s vs. 14s!)
