E 400 convertible
Won't go into gear

I have a new to me 2017 E400 convertible and occasionally the car displays a message of Driver assistance inoperable and Collision avoidance warning. I have also had an issue with the rear tire losing air slowly. If I don't stay on top of the slow leak I get these driver assistance warnings, tells me to refer to the Owners Manual (which is of no help)and the car will not go into gear. Fill the tire to 45 psi, wait overnight and the car magically starts and goes into gear. Can it be that simple that these warnings are related to TPMS ?

Have you tried fixing the slow leak? I have never heard of low tire pressure being tied to the drivers assistance package, but who knows. If you just bought the car, I’d go back to whoever you bought it from.

2018 E400 Coupe
Doubtful. The car (or at least my 2018 E400 Coupe) will start with a flat tire (although it does warn me about the flat).

I strongly suggest you stop driving that car and have it taken to the nearest dealer or repair shop that specializes in Mercedes ASAP.
