What is the main diff b/t 08 and 07 GL450?! Should I go with an 07 for that few $K saving?!

P1 pkg is different in 07 vs 08, in 08 rear view camera (and some other items, don't remember) comes with P1 pkg and in 07 you have to pay extra. I think P1 pkg in 07 is $4500 while 08 is $6500. There is no difference as far as engineering and handling between the two model.

Put-it-in Park chirp went away for 2008 IIRC.

Also IIRC change in headlight wash switch.

The tan/beige/gold metallic paint was changed for 2008.

Go with the 2nd year version always! Not very scientific, but it always seems to be a good rule of thumb. Mine runs pretty flawlessly. Good Luck!

I thought 07s came with a full size spare and runflats became standard starting 08 and no spare.

Runflats are standard on 09+ Bluetec diesels beause the spare tire bay is occupied by the AdBlue tank. The 07/08 should have 4 normal tires plus a spare.

'07 and '08 have "spacesaver" spare and no room for a full size spare.

(If you live in Europe you can get a tailgate mount for full size spare, but that's basically impossible in USA.)

Runflats and no spare at all began in '09 with advent of AdBlue.

Is there a mileage limit running on the spare because of the size difference before burning up the AWD drivetrain differential?

Its more a matter of the "donut" spare's size that you are supposed to only run about 50MPH or it will blow apart. It is not meant to run for a long time, just to get you to a service center. That being said, I have a friend that ran the donut spare on a E350 for two weeks as the dealer had to order a new rim, so it took that long. (I know the E350 is not a GL 4matic but just letting you know the spare will usually go a while)
I dont believe there is any issue with the differential.

There may not technically be an issue with the differential but when I tried to find a replacement for one of my tires a couple of years ago (Discount Tire road hazard warranty replacement) and I had to run on the donut for a while I both felt and heard the difference in the tire. I believe the total circumference of the donut is different.

True. I didn't mean there wasn't a difference in the donut size (yes it is smaller). I meant that it would not harm your differential. Even if you had a positive traction rear end in the GL (which we don't), as you go around a corner the inner tire would try to spin as the outer tire is spinning faster causing a hopping or skidding noise. If that is the case, the tires would wear prematurely every time you go around a corner. That being said, that would happen even if the tires are the same size or not. If it were me, I would not be comfortable running the spare for any length of time due to its small size and potential for blowout and subsequent saftey issues, not for mechanical issues of the differential.
