In the GLS, C. The S mode lowers it a bit, stiffens it up a bit but MOSTLY...you start in first not second gear. In the 350 S, in the AMG S (S+ from time to time). In the C, no difference I can tell....
In the GLE 350, eco mode made more sense and it's still second nature to me as soon as I start moving to switch out of comfort to eco. Now with the 450, I don't know if it matters. Why do you say comfort mode makes the most sense?
Time to use individual then : ) and set it as desired.

Do you have the air suspension? Sports mode will significantly tighten up the suspension with AirMatic. I find that Eco mode is not that useful if you use the cruise control. It won't shut the motor down and coast if the cruise control is enabled. For daily use and highway cruising, I use the Comfort mode and turn off the stop/start. For sportier roads like the Blue Ridge Parkway, I put the car in sports mode.
I never use sport mode. My preference is eco mode because the torque converter unlocks and allows the car to coast. I suppose this bypasses some of the regenerative braking that puts a charge back in the 48 volt battery but that's the only disadvantage I see. My car has the steel spring suspension so that's not a factor. I don't think there's a right or wrong and it's good MB gives us some options.

MB couldn't care less. MB gas economy and emissions are measured in the lab or on track in a standardized format. The different modes are to satisfy as many owners as possible. Different people have different preferences and priorities.
Just zipping around town running errands: Sport mode
Commuting to work (55 miles each way): Eco but I have been using Comfort the past week to see how much of a difference it makes. (Doesn't seem like it makes a huge difference, eco gets me a couple more mpg, if that)
Road Trips: Eco (and switch into Sport if I need a power boost like passing someone or something).
I'm in comfort mode most of the time. I would say that I never use Individual, Sport, or Offroad modes--even while off road, I still use comfort. I use Curve for canyons, winding roads, freeway on ramps; Eco on road trips if the road is smooth--I do enjoy seeing the green D to show that I'm sailing; and Sport + during spirited driving.

Yeah, I'm surprised as well. My 2017 GLE 350 had a Snow/Slippery driving mode. There was one time this winter we were getting heavy snow in my area (roads are rural and twisty) and I put my MY24 450 in Off Road drive mode. I had to drive slow anyway and figured that would give me more traction than say Comfort. I could be wrong though. ¯\_(ツ)_/¯

There is something called all weather tires for those who hate seasonal changes.
