When I start the car, is it possible to :
- Lock the Sport Mode to keep it in Sport Mode for the first mode when I start the car ?
- Put to Digital Speedometer by default when I start the car.
(My wife C300, when we put the digital speedometer, it's always stay when we start the car until we swith for other thing, but seem not stay for the GLE).

default is factory initial setting, what will happen in case your battery gets disconnected.
The screen view should remember last setting on other hand and I often use it as a reminder for next day to inflate my tires when they go low (I inflate them at cold for proper readings).
But MB plays with software without notifying the owners, so if your MY have it changed - it should not be a surprise.
For #1 I recall Russian seller on ebay offering OBD plugs who will override factory software. Not cheap.

I'm sure there is a reason why C (Comfort) drive mode is set as a default . I think it's have to do with the CARB (Emission)
My old W209 will keep what ever mode you were in saved so next time I start the motor up it's already set to that drive mode.
I'm wondering if you can program it through STAR / DAS to set the 'S' drive mode as default
