Recently purchased a 2014 GLK250 with the same shudder when going uphill over 60. Torque converter replaced under CPO warranty. Vibration gone and drives like new! Couldn't be happier with this car.

2015 GL550
Had the exact same issue with my 2015 GL550 (~65K mi / 104K km). Turned out to be just a failed ignition coil (#2 to be exact).

Did the failed ignition coil eliminate the shudder on acceleration completely?

It has. As soon as the failed coil was replaced, the vehicle was back to normal.
