Will these fit my s500
I have an 03 s500 will these fit-Size: 20x8.5

Depends more on the tires size you put on. I would suspect the w/o rolling the rear fendors, plus cutting the plastic parts. It won't fit w/o rubbing in the wheel wells, if the car is fully loaded and on bumpy roads, or at high speed (car lowered) on the highway hitting a pothole.
A good test to find out is mounting the thing, drive on a pedestal (or ramp) of ~6-7 inches with one rear wheel and turn the steering wheel to the opposite side where the elevated rear wheel is. Sit 2 persons in the truck and see what you have. This is about the maximum (worst case) to expect. I did this and for the W220, the unexpected was that the wheel on the ramp is moving not only upwards but also towards the rear end of the car (given by the geometry of the suspension) This in most cases requires to cut parts of the plastic of the bumper and rolling the fendors.

I actually went with a different wheel, "MHT Stoke" 20 inch. They are custom built for the car and are very high quality. Will I have any problems attaching the tire pressure monitoring system?

I guess, yes. You need to find suitable space to attach it. I don't know how those sensors are attached originally and if they require calibration, after being of and mounted in a different wheel.
