You are in the wrong place - this is the W220 Forum (S Class, 1999 - 2006). Your 2000 SLK230 is an R170, and there should be a forum for those.
I can tell you what the problem is likely to be on a W220, and the check might be the same for the R170. First thing to check is to turn the ignition switch on, hold the brake pedal, and look at the the instrument cluster. In the W220, there should be two letters there - one should be "P", and the other one will vary, depending upon what your particular car calls the two selectable transmission shift patterns (might be "W" for winter, "C" for comfort, "S" for standard, etc). While still holding the brake, slowly move the shift lever to the reverse position - the "P" should change to "R". Move the lever to the neutral position, and the "R" should change to "N". Move to drive, and the "N" should change to "D".
If none of this happens, the rod linking the shift lever to the transmission has fallen off at one end. There is a different plastic bushing in the end of each lever. They are available at the dealership for not too much money. Installation isn't too difficult, but you do have to SAFELY jack and support the car for access. Even if your car doesn't have the letters in the cluster, the problem and fix are still very likely to be the same.
