Replacement Tires - higher aspect ratio
I have a 2012 S350 Blu-Tec with Continental 255/40 R19 100V tires and I hate the ride of these low profile tires, Is it possible to go to a 45 or 50 aspect tire with this vehicle without causing any other problems ??

You would want to maintain the same sidewall height to maintain overall tire size. The formula is 255 x 40% = 102 mm for your current tire. If you went with a 45 series tire, you would need to find a 225/45R19 tire (225 x 45% = 101.25 mm) or a 50-series tire would need to be 205/50R19 (205 x 50% = 102.5 mm). I'm not saying these tire sizes exist, only that they would give the same overall tire height as your current, albeit with a narrower tread width. The 225 tire is 30 mm narrower than the 255, and a 205 tire is 50 mm narrower than 255.
