Hopefully I can describe this since I can’t get a decent picture... the owners manual mentions a square area to the right of the rear view mirror for toll tag transponders. I also noticed a much smaller oblong opening in the center of the windshield under the cameras.

Is this opening for the smaller oblong toll tags or would the transponder interfere with something?

You can use either one, both intended for the same reason.

Yes that is where you put the toll pass and radar detectors. If you stand in front of the car wearing polarized sunglasses you can see it clearly. Otherwise the light and angle need to be right. On the 2016 S550 it is center down near the dash not near the cameras.
