It just arrived today. I have a few things to do before I will plug it in. Spark plugs, exhaust flap issue, engine and transmission module updates need to be addressed before I do anything with the tune. In the meantime I'm playing with the lowering module that came with it. I will start a new thread or continue another one on the engine tune when it's time.

The Cylinder deactivation is actually very hard to feel. It was done well by MB and most would not notice it. I think in reality what I'm feeling on the highway is tire vibration. Either my wheel balancing is off or the run flat tires are producing a slight vibration or it's possible I have a slightly bent rim or two. You can feel the cylinder deactivation if you are cruising along say around 50 mph under light throttle load and then you push the pedal down a little that slight vibration will go away as all eight cylinders are activated. A lot of people might not even be able to pick this up as it is so faint. I guess I'm just really perceptive with mechanical type things and feel these different things going on. Gonna bring the car in this week to get the wheel balancing checked out, I don't know why I'm even screwing around with this and not just scrapping the run flat tires. The tires were brand new when the car was Certified 3 weeks ago and only have around 500 miles on them. Conti SSR run flats. Hopefully I can make them work if only they would be a little smoother on the highway, I sure do hope it's just simple wheel balancing.
I can attest that it's NOT the tires. I got rid of the (awful) Goodyear run-flat tires and put on super quiet and comfortable riding Michelin Primacy Tour A/S tires which are worth every penny in as far as ride comfort - they put the ride on par with the Rolls-Royce Ghost. (I keep a tire repair kit and a compressor in the trunk.)
I have a 2008 Chrysler 300C with the 5.7L Hemi engine and I noted this same subtle vibration phenomena when half of the cylinders deactivate. I had been running regular (87 octane) fuel in it and I noted that the vibration is more pronounced, so I switched back to premium fuel. It's still there, albeit a bit less.
So now, this all makes sense! - the S560 vibration is very reminiscient of the Chrysler although less pronounced.

I notice the vibration quite a bit but always try to remember to turn the eco button off. It’s hard on the engine too running on 4 cylinders. I’ve still got the Conti ssr tires on the car 50 k miles later. They have surprised me how long they have lasted. I think I’ve got at least 10k mikes left on them. The next tires I get though will probably be the Conti Dws o6 plus non run flat tires. They are less money than these run flats and the car will ride better.

I test drove dozens of W222 facelift vehicles before I bought my last W222. I can tell you that in almost everyone one of them I felt that harmonic vibration because of the 4-cylinder deactivation. I got that taken care of by asking @EleBest to code the vehicle and problem was gone.

Could be motor mounts. Had one S Coupe and one CL coupe vibrating. New mounts corrected the vibration.

Peter AKA BenzNinja can turn off start stop, and a nickel in a bet says cylinder deactivation can also be asked to not operate as well.
I had had him give my car ‘remember last mode’ such that it wakes up in sport mode... no start stop.
Many excellent options that can be enabled while annoying ‘features’ can be turned off via software.

Only issue with the remember last mode on a refresh W222 is it only remembers it for about 4 hours. Still a huge improvement but every morning I still have to change it, but when it gets restarted throughout the day it still remembers. Just a quirk of the new system.

There is a video on Youtube on Mercedes cylinder deactivation, Also if you download the AMG S560 manual, on page 28 there is mention for AMG Models with cylinder activation, but not much information on how it works, just how to enter it and how a symbol on the instrument cluster will show how many cylinders are activated. There is NO mention of this in the manual for a non-S560 and probably for a good reason imho. I own a '19 S560 sedan and had a S560 Coupe and frankly am a non believer in both those cars. LIke I said, Non AMG cars.

Just out of curiosity is there a Icon on the display of the S63 models that indicates when it changes to 4 cylinders? I know on my 2018 E63 amg there was but no such light on my 2018 S560 but it definitely has it. I figure Benz didn't include the Icon on the regular S560's as they figure people driving a standard S class are not as likely to be tech savvy as someone driving a AMG car, just a theory lol.

From MBUSA Download the 2019 manual for AMG Owners Manual and on page 28 it shows the symbols of where and when the AMG changes from 8 to 4 cylinders. Not to be found on a regualar non S Model Sedan and Maybach, anywhere to my knowledge.
On AMG models it says you have to be in Drive Program 'C' and have the ECO button on. So I assume if your in any drive mode other than C it will not deactivate 4 cyliiders.
cAdditional notes regarding your Mercedes-AMG vehicle:
Vehicles with an Instrument Display (standard) only: If you select the Perform- ance menu, the upshift bar appears on the multifunction display.
IfyouswitchontheECOstart/stopfunction and select drive program A, the engine electronics switch from 8-cylinder mode to 4- cylinder mode, if necessary. The multifunc‐ tion display shows the / symbol in the status area.
