If we are still discussing W212 E63 S model's , they are wet clutch transmissions that aren't equipped with a torque converter .

Ive had this exact issue on my S212 2014 facelift. It was right after i changed the plugs, it ended up being a loose connection between the wire harness and the coil pack after i had done the work. HOWEVER i did throw CEL and was able to find the cylinder that was misfiring and corrected the connection. No issues since. I'd double check the connections, i did the plug change and had no issue for 2 days, then did 400+ miles in a day with thruway as well as on some very rough roads, after the rough roads is when it started, im pretty sure it just shook loose because i may or may not have broken the stupid tiny dumb gray clips.... nothing some wireloom tape doesnt fix.
I'd say check the connections to the harness and the boot, as well as just make sure your boots are good on the coil pack, but everything about this looks like misfire, i can't really tell by the audio in the video, but do you notice any change to the engine noise other than just load?
