Engine software update
Had my service A done recently and was told there was a "recall" / service campaign from MB for my wagon (2016 E63S). Campaign #2017090009.
Overall the throttle inputs feel a little smoother in each mode but that might be me imagining things. One thing I noticed is that the car only starts in 2nd gear in comfort mode without exception. I honestly don't recall that being the case before - 2nd gear when cold but not really when warm. Can anybody confirm on their cars? Doesn't bother me (unless I'm losing my mind which is possible...) just curious.

and seems that the car always starts up in C mode, then of course you can adjust the trans manually turning the knob or using your AMG button pre-set assuming its set to S or S+

Yes, the M157 is default to start in C and the default can not be alter unless of course the turn that knob to sport or sport+.

You CAN start in 1st from C mode
In C mode all you need to do is pull your downshift paddle and you can start in 1 but still be in C. After a few shift it will return to normal 2 gear start.
Once you pull the paddle you will notice the cluster change from the drive letters to the 1-7 number of gears.
