an E250 wasn't sold in the USA, hence I'm guessing you're elsewhere in the world, and that means you could have any of about 5 different media centers. i'm personally only familiar with the NTG5.1 / Harman Kardon Logic7 system in my 2016 US model E350. I generally use Spotify on my phone rather than a USB stick, in fact I don't think I've messed with USB stick based music in about 10 years..
The COMAND system does have an option to play "random order". Perhaps not all models feature this option.

most things that play MP3 media files off a USB stick will also support a .M3U playlist. this is a simple text file, that can be created in any text editor such as notepad, one line per track with the filename... if you tell the media player to play that m3u file it should play the tracks in the order listed
now, I've never tried this with the Mercedes COMAND system, as I haven't messed with mp3 files or usb stick based music since long before I got the Mercedes. When I did use USB sticks, they typically had 100 or so folders on them, each folder being a complete album, as I usually like listening to albums rather than individual songs.
