Rubber housing allows bearing to move radially under different torque conditions and when you put your tranny to N the torque goes away and the bearing moves changing the angle at joint. This could be the reason why humming (vibration) stops.
There also are rubber flex disks at each end of the propeller shaft that over time develop cracks and can cause vibrations under torque.
Then, if the tranny mount sags it also changes the Propeller shaft angle and could lead to vibrations.
I would check the tranny mount first, flex disks second and then the propeller shaft center bearing. This bearing probably is fine but the rubber housing is not. I changed this bearing on my S550. The rubber housing had sagged about 1/8”. I think E-class has the same design.

WRT changing the tranny fluid, I would do that regardless if you don't know when it was last changed. Depending on whether you have the red or blue fluid (THEY ARE NOT INTERCHANGABLE!!!), you may be long overdue for a transmission service. Red fluid (ATF-134 or MB 236.14) should be changed around 40,000 miles and blue fluid (ATF-134FE or MB 236.15) should be changed around 70,000 miles. Figure around $600-$700 at the dealer, but a lot cheaper than a new transmission.

One way to make sure is ask the dealer. Just give them the VIN and they will know. Also, your service manual should say what the fluid change interval is. 40 000 means red and 70 000 means blue like JettaRed already mentioned.
