From what I understand, tuning these cars may cause the coil packs to fail earlier than normal (that was certainly the case with the V12 motors) and perhaps the spark plugs may need to be changed earlier than what MB recommends. But that is a small trade-off for unlocking the potential in these motors. Your V8 450 will feel like it got a big injection of steroids as you can get another 180 hp and 200 lb/ft of torque just from the tune and nothing else. Also better gas mileage if you don't have your foot to the floor all the time. It's great that you can get them now with the hand-held tuner that you can easily put back to stock any time you want. That wasn't an option when I got mine.
Another thing the tune may cause is for you to need to change your tires and brakes more often. Mine will spin all 4 wheels on dry roads from a dig, so the tires wear out faster and the ABS intervenes a lot more often causing more brake wear. Fortunately it's still possible to drive these just like most people and have no extra brake or tire wear.
