I'm getting new shoes for my GL550 and I'm going to try the Conti ExtremeContact DWS-06s. Tirerack has them for $322 but they are available locally (DFW) at similar prices without having to wait for shipping.

The 295/40/21 DWS is new. Pirelli Scorpion wore out about 30k miles on me, and are very loud. Good tire when new, but wear out faster than I would like and also don't wear very well.
UHP is the second best all-season for this size right now and is a good tire.
DWS I always purchase if available, so pleased it's in this size. It's an easy decision for me for those that need all-season, not to mention it has a 560 AA A rating, the best tread wear ratings available. I have not tired it in this size, but have tried in in 2 other sizes 17" and 20" and it is now my go-to tire. Note I run Pirelli Scorpion Winter for the winter, but all-seasons are great in case I get a little lazy in switching over. Summer tires are way to slick and would have to switch them immediately once temps average below 45 F.
Looking forward to the 295/40/21 DWS review, but I suspect they should be consistent with the other reviews of DWS tire in general.

I've had a chance to put a few hundred miles on the Conti ExtremeContact DWS-06s. I am delighted with this tire. Great handling, quiet on the freeway and great in the wet, even during the torrential downpours we've had in Texas lately.
There isn't anything I dislike about this tire and I would definitely recommend them.
Can't comment on the wear, obviously, but so far, so good!

I have a 13' ml550 with 275/45-20 conti DWSs. Great performing tire but I'm almost to the wear bars in 17K miles. Replaced the stock contis with them. I drive aggressively but got 30k out of the stock tires. Great tire performance wise but tread wear is nowhere near what they state. I run Pirelli ice and snow in the winter, same size.

I'm very disappointed with the UHP's on my GL550. We have not been hard on them at all and a good portion of their 23,000 miles was highway. They did wear evenly but would not stay in balance and became very noisy after 15,000 miles. I wish I had more than 7 choices for tires in this size. Based on what I've been reading I'm going to try the ExtremeContact DWS06. They seem like my best choice. I'm still a little nervous because the one GL owner review for this tire on tire rack stated that this was "the worst tires he ever owned".

Had extreme contacts times 2 on my x164 GL They were actually quite good. I had no issues. I a have also had them on other cars with no issues. Stay away from Pirelli.
