Just received our 2021 GLE53 SUV last night and performed a manual Map/GPS update via the Mercedes Me Connect website. I used an ADATA 32GB USB Thumb Drive, formatted NTFS, partition style MBR. Here's the steps I went through, worked flawlessly.
1. Using Firefox on Windows 10 PC, selected a few different map regions to update (about 10GB worth). Downloaded the Mercedes Me Download Manager (option provided on the site) since this was my first time. After installing the MMDM, files downloaded and copied to USB drive without any issues.
2. Used a USB-C to A adapter I had laying around the house (I actually received two new ones with the car/dealership), connected to the USB-C port next to wireless charging pad in the center console. The MBUX system automatically alerted with a pop-up (Photo 1) informing of a Software Update Available. Tap
3. Tapped the available Map Update, North American 10GB (Photo 2)
4. Tapped Update (Photo 3)
5. Install began, took ~20 minutes or so (didn't time it exactly so guesstimate) (Photo 4)
6. Once completed, MBUX alerts and requests a Restart, tap (Photo 5)
7. After restart, MBUX informs Successfully Installed (Photo 6).

Note: after starting the update process while parked, I was able to drive while it performed the update without any issues. I left it on the Updating screen and didn't use any functions during the drive.
