2016 GLE350 rough drive and idle

This weekend my CEL came on and then I noticed what seemed to be a tire with a knot in it giving a light thump as we drove down the highway. We pulled into the parking lot when we arrived and as I slowed down to park the car began running very rough like it was missing. I checked the tires and they all looked great (less then 6 months on them). When we got back in about 30 minutes later the CEL was off and it drove great. Yesterday (2 days later) as my wife was leaving work the issue reoccurred and as she was in traffic the rough low speed issue was much more pronounced. When I checked the codes with my reader it shows a Fault with the communication on the VGS Transmission Control. Has anyone had this issue, or know what I am looking at to fix it? Only one MB dealer in over 100 miles and they cannot look at it for over two weeks.

Misfire.

Possible reasons:
- old/bad spark plugs
- old/bad coils
- oil in wiring harness from leaking cam position sensors and cam magnets
- bad injector (unlikely)
- failing purge solenoid
- failing PCV system
- intermittent intake leak (cracked purge or PCV hose, leaking intake manifold gaskets)

Read codes and post them here. Autozone guys can read codes for free.

When were the main and auxiliary batteries last replaced?

Both batteries replaced in last 6 months, 7000 miles. Not sure when plugs were replaced, follow the recommended A,B,... service. Have a reader, will read active code when I get home and post them.

P035613 - Coil 6 electrical fauly/open circuit
P030685 - Combustion misfiring of cylinder 6
P030085 - Combustion misfiring detected

Well done. How do the connectors look at the cam position sensors and cam magnets? Any signs of oil present when you open the connectors?

I move the Coil pack from cylinder 6 to another cylinder. If no CEL shows up it's not the coil pack because if it was it be on a different cylinder and it be a different code. So the P030085 would be cleared.

Update - swapped the #6 coil pack with the #4 one and the problem followed the coil. Replaced all six coils and the plugs as a set due to age. Engine codes have not returned and it is running smooth. Have slight seepage from left cam magnet so will be replacing those soon. Thanks for all the help!

Welll done. Coil replacement is a very good, arguably necessary, maintenance item at higher mileage. I changed them at 100k miles, sooner wouldn't hurt. Later would invite CEL and rough running.
