Money no object, who do you have tune your E55?
Next step down would be stand alone ecu and get somebody who really know how tune that particular system. Better yet, have him build your car, install the stand alone ECU, and tune the it. That way there will be no suprise.
Another step down from there? Depend on where you are, get a tuner that is best with MB in your area and get to know him on a first name basis. Why? Because you will need him down the road. I like to deal with people within driving distant.

Course if money is more of tiny little object. Heard nothing but great stuff about Jeremy so far. Renntech is also getting a bit more aggresive with their tunes and mods. Never would have suggested them in the past but after Benzgals car and couple others, they are finally getting a bit more radical with the tunes.

To answer the "why?": performance, exclusivity, history/reputation/prestige, reliability, resale, depth of engineering & manufacturing capabilities, and comprehensive turn-key solutions.
Reason(s) not to: cost. Period.
IMHO, Renntech and Kleemann are excellent tuning organizations, and would be my "costs-considered" choices, with Kleemann being the better value proposition these days.
The current 5-speed maybe more proven for those high hp/torque applications as repeatedly illustrated by the V12 monsters.
Otherwise I love my new numbers from Tony @ let/eurocharged
