help roof won't work on my 2000 clk430

The opening is to the left of and slightly lower than a similar flap, used to release the hydraulic pump relief screw, that is used to raise the top manually (see owners manual).
Sounds like you haven't used the forum's search function.
The power roof is very complex and it's operation involves an electronic controller, hydraulic pump/motor/relay, 7 hydraulic cylinders, 15 or more limit/position switches, tension cables, windows and roll bar operating properly.
The W208 convertibles are now 12 - 16 years old so, it's not a question of 'if', but 'when', something in the power roof system will require repair.
As a result, if you have good trouble-shooting/repair skills. there is a ton existing DIY information in the forum (e.g. search is your friend).
I know it's a pain, with many dead ends due, due to numerous new members asking for help on a topic that has been already, throughly covered.
Alternatively, the STAR diagnostic software, available at MB dealers and some independent mechanics, can be used to further diagnose and/or repair the problem.
welcome to the forum! Where are you located?
It sounds like you will have to roll up your sleeves a little for troubleshooting, or pay a dealer for reading codes via StarDiagnostics. If you are willing to roll up your sleeves, after doing some reading, then I would recommend browsing through this most comprehensive thread on the subject: https://mbworld.org/forums/clk-class...-failures.html
Checking the fluid level in the pump is essential (as suggested by serndipity), as are checking out the 40A fuse in the trunk and the relay next to the pump. Instructions for access to the pump and the relay are part of the following DIY: http://www.tophydraulicsinc.com/W208...eplacement.pdf
