My wife took the car the other day and said she had problems locking and unlocking the doors. She also said she got a message on the console stating Key Not Detected and the car Keyless Go wouldn't work. I thought it was the battery on the key fob but then remembered I had just changed both of them. I thought the issue was isolated to her fob but then it happened to me yesterday. I called they dealership and told my SA what was happening- he didn't sound surprised. I told him I was aware of the Static Electricity theory and disproved it by telling him my key was covered and I tried a Bounce Sheet in my pocket. I'm having the curb rash fixed on my wheels and Cilajet applied next week so they said they'd take care of it then.

My wife had her key fail on vacation. She went to the store and came out and the car would not start. Had to be flatbedded 75 miles to the nearest dealer in Phoenix. She rented a car in the town the key failed. Warranty paid for everything but it was a PIA. She now keeps the second key in her trunk because they need it to diagnose that a key has gone bad.
Did some searching and there are 3 known issues:
1. EIS
2. Keyless Go antennae in the driver's side door (defective wire harness)
3. Key Fob Battery (Already Replaced)

as far as problem #2 i read on S class forum that there was a DIY on how to fix it. there is a wire that breaks off over time or something.

#3 did you buy the battery yourself somewhere or did you buy from the dealer? My dealer had a bunch of bad batteries. I bought at a store and replaced and it worked fine after that.
Try taking out the START button from the ignition and use your key fob manually. I was told somewhere by someone that on rare occasions the key fob can dissociate itself from the Keyless Go ignition and it has to re-learn the fob.

That is the only way I could start the car. Today, I stopped for dinner with my wife. When we returned to the car, I was unable to unlock the car by the Keyless Go OR by the Keyfob. I had to use the Valet Key to unlock the door and then the Alarm went off. I had to jump into the car, pop out the START button, insert the key and start the car for the alarm to turn off. Considering I'm scheduled for Back/Neck surgery, and just had a Heart Catheterization which didn't go too well within the past week, this made me VERY UNHAPPY!

I'm surprised that the car will lock with the other key in the trunk.
No problem with car locking. I don't have keyless go

Not having the second key added 3 days to the repair because they had to get a new key from MB to diagnose the problem. Their ignition diagnostic REQUIRES both keys to run. Had she had the second key, she could have used it and avoided the flatbed ordeal.
Just got back from the dealer. I too was experiencing many "key not detected"
issues. I bet at least 10 a week. Tried new batteries and switched fobs with my wife even. Mentioned it to the dealer and was told that I had an Energizer battery in the fob and that brand does not work well with the system. They replaced it with a new battery that is made by Panasonic. I will keep track going forward. Not sure if it is BS or not but, I have not heard of the battery manufacture as an issue before.
Did they charge you $10 per battery? There is something seriously wrong with the sensitivity if Brand of the battery can affect the operation of a keyfob.
I had both keys today since they said they needed them to troubleshoot. Had no issues at all. I told the SA and He laughed. He asked me to forward him the pictures of the Red Screen and said they would thoroughly check everything.

I had the same issue a few months back. I had previously replaced the fob battery with a new Duracel part after low battery warning. Soon after, same red warning. Took it to the dealer, they replaced the battery with a MB part (Panasonic) battery, and the issue has not come back. Very strange. No charge under warranty.
