They had my truck in the shop for 14 days total on 3 different occasions trying to fix problem. Last time they said they put someone on the seal and it did help but after three weeks noise has returned. I am going to schedule another appt. The noise irritates me more than running your nails down the chalkboard.

Any update on this? Did you bring it back to the dealership for another fix. I drove mine again this weekend and heard the whistling each time I hit 65mph and above. So ridiculous on a luxury SUV. My 1992 jeep didn't even do this! Getting more and more aggravated with this SUV.
The only real way to fix this is to have a tech take your car out several times out on the highway, and place a microphone in various strategic areas beforehand in order to narrow down the source of the whistling. It will remove the guesswork.
I have a GLE 580 that I bought 2 weeks ago and the same issue with whistle noise on the driver side once achieve 60 MPH.
I have not talked to the dealer about it yet.

Went on a long drive (Boston to Washington DC and back) and it happened to me a dozen times (and captured it on video too).

No, that is not the noise I have been describing on my GLE. The whistling I hear is much more subtle and is coming from outside the driver's window. If you had music on, you would not hear it. However, it is one of those things that once you do hear it, you hear it all the time and listen for it...very annoying.
So today I did a quick test, folding the mirrors the noise disappeared, and when I opened it back the noise came back, so it is definitely related to the mirror.
This weekend I will try masking the seams on the mirror to try to find the exact location of this noise.
Did anyone tried this test before?
I cannot handle this whistle.
Do any of you happen to have protective film on the mirrors or A-arms by chance? I once had a volvo s60 and the film was the cause of the noise at high speed???
Actually, we have protective film on the rear view mirrors and on the A pillars. I'll chat with the dealer to see what they think. It's been on since shortly after we bought the vehicle, and it's been on there every time they've looked at it, so I would hope they would have thought of this, but I'll definitely talk to them about it.
I have to take it in soon, anyway, because the auto-dimming headlights don't work properly, and turn the brights back on randomly as oncoming cars approach. So yay... more time in the shop.

the problem is the side mirror cap.
i had to add 3 layers of electrical tape between the frame and the cap, all around it.

if anyone finds out from Mercedes what will be their resolution for this, please keep us updated.
Maybe they have to redesign the mirror to correct this issue. Nice job figuring this out. Hopefully the other people who have had this problem will try your fix to see if they have similar results. Mine is still on order, so I can't give it a try yet.

It's odd that we haven't noticed the issue at all, and I've driven up to 100 mph.
If the gap in question is the gap between the mirror and the mirror housing, then maybe it depends on the individual mirror angle position. If that's the case then some may experience this whistle while some may not. Just a guess.

That's an easy experiment for those with the whistle. Next time you hear it, adjust the mirror and see if it stops.

Interesting. Sounds like there are a couple different whistling gremlins in the 2020 GLE.
One is from outside the drivers side window and another is from the drivers side windshield.
Frustrating part is that it doesn't happen all the time and when you report it to M-B service, the technicians are never able to reproduce the noise.
At least there is video I can show them. Hopefully capturing it on video can help identify the source.

I am going to go in this week and tell them I need this fixed. The noise has come back so whatever they did was some type of temporary "fix." I was told by the service manager that a Mercedes service engineer told them over the phone how to "correct" the issue which involved something on the driver side rearview mirror seal and some type of added adhesive. First, the noise never went totally away when I test drove it, but it was dampened. Now, the noise is back in full force. I think owners are going to have to make a big fuss over this until Mercedes does something to fix.I put a rag in between the rearview mirror and car bod and the noise is gone when going down the highway. I am about to record a video and post online showing this and saying "this is what I have to have to do have a quiet ride in a $70K vehicle." If enough of us do this and post then I have a feeling Mercedes will respond.

This fixed this issue for me! I too now have electrical tape on my $70k plus car so it does not whistle as I drive? Does anyone know if replacing the side mirror fixes the problem?

I would think clear silicone sealant in the crack between the mirror housing pieces might be as effective and look better?
Reminder, it is not the mirror itself, it is between the frame and the cap cover.
The mirror has nothing to do with it.
Interesting. It appears that high pressure air (from being pushed around the side mirror) is being sucked through a gap/crack between the frame and cap cover, to the low pressure side on the face of the mirror (there is a vacuum on the face of the mirror and the space between the mirror edge and inside edge of the cap cover). The whistle sound must be a result of air being sucked through the gap/crack and out around the edges of the mirror glass. To stop the whistle, you have the stop the air from being sucked through the gap -- my guess is that's what you're doing with the electrical tape. I'm not familiar with the design, I wonder if there is a rubber/silicone gasket between the mirror frame and cap cover that is not seated correctly (or is missing) that is intended to prevent this? I don't have the whistle on my vehicle.
