GLS 550 Transmission & Spark Plug Services
I just received quote of $850 for Transmission service and $720 for spark plug change. Are these prices out of line?

Not too far out of line. $600 or so was normal for transmission service before current inflation. Spark plug change on the V8 seems in the ballpark. You might be able to shave $100 off each job at an Indy if the prices you listed are from a dealer.

Whats the mileage of the vehicle? I recently did the spark plugs at it was about $550. I had the ability to shop around and it ranged from $550 to $800 depending on dealership. I also had them match the price of the other dealership. Hope this helps.

I ended up needing add'l work done in addition to plugs and transmission service.
An extended warranty will cover CV/Axcel boot and engine mount replacements.
I would think there would be an overlap in labor between replacing motor mounts and spark plugs, but I was told there wasn't much. Thoughts?

It's a rip-off-the-warranty-company-scheme. Scummy dealers (all of them) will dip their hands into the honey pot (the warranty) as deep and as often as possible. It's a racket.

Not out of line. Transmission service involves dropping the 4MATIC support brace which is a PITA. The spark plug service is much easier and is 1 1/2 hours at home DIY. Might as well swap out the ignition coils while you're there because in my experience, Mercedes ignition coils can go bad after 60K miles anyway. Spending money up front is well worth the headache of a cylinder misfire.

EDIT: original post was corrected as it (referencing X166 GL's 7 speed) does not apply to X166 GLS ' 9 speed that was used on all models except 63.

Is that what a dealer does?
This is a video for W166 GLE63 and no support brace is removed: https://www.youtube.com/watch?v=5eIq_qGyw1Q
On my W166 ML63, dealer charged $555 (3 years ago) of which labor portion was only $235. I think they just remove transmission cover and then use appropriate tool to reach those bolts, just like the guy in the video.

it is possible to do a GLS without dropping the brace that has the transmission mount but you need to squeeze in tight with a tiny and high quality (tight tolerance) (think SNAP-ON) 1/4 inch 8mm socket to get at the E12 bolt above the transmission mount and rotate out manually most of the way or have a extra long 8mm ratcheting wrench to help. Getting the new pan back on is also a PITA and may require clipping in the reused plastic piece on the drivers side blindly by feel after the pan slides into place most of the way.
It is doable but not straight forward. I’ve done it and I was cussing thinking I was screwed half way through. I thought of dropping the transmission mount cross brace. But I worked my way out of it. The above video is for a 7 speed tranny. The 9 speed is a different shape and position.
If I didn’t have a huge toolbox (I have four full sets of metric ratcheting wrenches) and many sets of sockets, and didn’t already have a tranny fill pump, I’d definitely sent it to the mechanic for a tranny service.

Misfire, rough idle, poor fuel consumption, check engine light.
Potentially collateral damage from excess vibrations such as engine mounts, cracked plastic parts, etc.
No-start in extreme cases.

It's even easier if you remove the two 16mm nuts from the transmission mount on the cross brace and jack up the transfer case a few inches, its makes a world of difference getting at the rear middle pan bolt and removing the pan too.
