What's the best way to start rolling race?

What's the best way to start rolling race?
Only way i can think of is in manual but then probably won't shift at the optimum rpm.

2014 E63S; AMS 100 octane ECU dyno tune; EDOK TCU tune; BB intakes; sprintbooster
Be in lowest gear possible while in Manual and switch into s+ as soon as you take off

2014 Mercedes E63S AMG
You can be in S+ and use the paddle to downshift to optimal gear. It will upshift itself still since its in S+ and not M.

S+ like everyone said, then right before you mash the gas, press and HOLD the downshift paddle, it will downshift into the appropriate gear for you. No idea if it will up shift though LOL (should)
