w212 shifting
Does the transmission in w212 start in second gear in economy and first in sport like my 2007 r171 in comfort and sport?

Nop, this can be a bit of a lottery.

Engine conditions can lead the TCU to shift gear 1-2-3 poorly.

Tranny should start off in 1st gear after a complete full stop, regardless of E/S mode.

The difference is astounding.

E-mode defaults to 1st gear
Starting in 2nd gear runs longer...

I had to go down this rabbit hole to get ECU-TCU to use gear 1-2-3 well.
My car used to feel like a bus at the red light... now it feels like a hummingbird. It was caused by the engine only.
The tranny does its best with what engine is doing -

The tranny banging is caused by the engine impossibility to coordinate for seemless shifts.

Don't temper with 722.9P tranny once it has good ATF service. That's all it needs.

What I noticed on my E400 is that while warming up it always starts from 1st gear regardless of transmission mode. After the warm-up period is completed or if the engine is already warm from a previous trip, it will start from 2nd gear in E and on 1st in S in flat surfaces.

And I agree with Cali, starting from 2nd gear makes the car feels sluggish and underpowered.

Yes, my pre-facelifts both start in 2nd gear in E mode and only use 1st gear in S mode, mine are '12 and '13 models.

On my 2014 E550, it starts in 1st in E mode until it warms up, and once warmed up it starts in 2nd.

That depends on engine.
My 4-cylinder diesel always starts on 1st.
So is GLE with the same engine.
