The Mercedes Me Connect App says it hasn't updated for 3 days and neither has the online website.
Has anyone had that problem and any suggestions on how to get the car to communicate?

Mine stops connecting if the 12 V battery is low on charge.

If the car is under cellular coverage (for the provider MBUSA is using), I'd visit the car, turn it on for a while and see if anything changes.
Last time I lost connection to the car I did not get any 12 V battery charge level warning but the app suggested this could be the case. Charged the 12 V battery and all was OK. Of course this is just on possibility out of many more.

Now that the app does not reach the car, meaning the car does not update it's status to Mercedes servers, you cannot know if the battery was OK when the car was switched off but is not any more. As I said earlier, the car should send a warning about low 12 V battery but at least I did not get any when my 12 V battery got low and I lost connectivity with the car. I was actually changing high voltage battery charging settings, the app did not see the change and lost all control. The first change of settings however reached the car when checked there. Just saying you cannot rely on the status the app shows if the car switched connectivity off to save power.
Now it would be nice if others could propose some other possible reasons for your issue!

I've observed a bug in the app that clears when the phone (not the app -- the whole phone) is restarted.
Everything is working now. Had to call Mercedes Me Connect and they set up a case. They told me to wait 24 hours. Within about 12 hours it was working again. Not sure what they did, but all is well now.
I did have one question, which I don't seem to remember from my previous s-class.
Regardless if the key is in the car or not, if I unlock the car through the App, it automatically locks itself back up after a very short period of time. Obviously, if I unlock the car with the key, it stays unlocked, but not if I unlock it through the App. I can watch the car open itself up through the App, and then it closes again.
Is this normal behavior. MB says it is, but I don't remember my s-class doing that.

Yes, that's normal. Should happen if you unlock with the fob as well. If the car is unlocked, but you don't open a door within 30 seconds then it locks again for security. That's been on cars with fobs for a while, not just MBs. My previous Audi did the same. It's primarily so that if you accidently unlock it with the fob in your pocket it doesn't stay unlocked.
